چکیده

In general, the factors important for protein crystallization are important for RNA crystallization, but there are additional factors that must be considered in RNA crystallization. Because most methods used to purify RNA denature the RNA, the annealing step is critical. The principal variables in the annealing are the temperature (40—70°C), incubation time at the highest temperature (1—10 minutes), cooling time (seconds to hours), buffer concentration and pH, and ionic composition (5—20 mM MgCl 2 ). Each annealing protocol could be subjected to the Sparse Matrix conditions. Alternatively, the annealing conditions can be optimized by monitoring the activity of the RNA or its chemical modification pattern 23 as conditions are varied. There are limitations to sampling the annealing condition. The buffers and ionic composition are partially determined by the Sparse Matrix. The annealing solutions (A.l—A.9) are usually made from desalted RNA stock solutions, but they can be made from solutions containing additional salts or polyamines. When considering the ionic composition of the solution,

How to use dynamic light scattering to improve the likelihood of growing macromolecular crystals.

Dynamic light scattering (DLS) has become one of the most useful diagnostic tools for crystallization. The main purpose of using DLS in crystal screening is to help the investigator understand the size distribution, stability, and aggregation state of macromolecules in solution. It can also be used to understand how experimental variables influence aggregation. With commercially available instr...

Entrapment–D-(+)-Glucose Water Nanodroplet: Synthesis and Dynamic Light Scattering

In the present research, the D-(+)-Glucose–confined water nanodroplets with mean size in the range of 6-11 nm were synthesized at D-(+)-Glucose different concentrations by AOT reverse micelles (RMs) method as a function of mass fraction of droplet (MFD) at the constant water-to-surfactant molar ratio ( W=40). The dynamic light scattering of the nano-sized water droplets containing the D-(+)-Glu...

Dielectrophoretic dynamic light-scattering (DDLS) spectroscopy.

Dielectrophoretic dynamic light-scattering (DDLS) spectroscopy is presented. DDLS identifies macromolecules based on their dielectric, or polarizability, properties. DDLS measurements are carried out in an oscillating, nonuniform electric field. The field induces macromolecules to undergo dielectrophoretic motion, which is detected by the modulation in the dynamic light-scattering autocorrelati...

Determination of the size distribution of monodesperse and bidisperse mixtures of spherical particles in the nanometer and submicron size range by applying cumulant analysis and contin algorithm in dynamic light scattering

Determination of particle size is one of the major needs in the industry and biotechnology. Dynamic light scattering (DLS) is a widely used technique for determining size distribution of spherical particle in nanometer and submicron size range. In this method, there are different algorithms for determining the size and size distribution of particles, which are selected according to the required...
